From San Francisco, you can reserve a ticket all the way to Yosemite through Amtrak:
You can also take a YARTS bus from Fresno, Merced, Mammoth Lakes, and Sonora directly to Yosemite:

Getting around Yosemite
Free Yosemite Valley shuttle system:
Free Yosemite Valley-Tuolumne Meadows Shuttle
The bus travels between Yosemite Valley and Tuolumne Meadows Visitor Center, stopping at various trailheads on the Tioga Road. Service is available from mid-May to September (see schedule here).
Leaving Whitney Portal
Hikers generally hitchhike from Whitney Portal to Lone Pine if they don’t have a car or a person picking them up. With so many day hikers going to Mt. Whitney, it’s generally pretty easy to get a ride into town. From Lone Pine, you can take an Eastern Sierra Transit bus to Reno, Mammoth Lakes, or Lancaster.
